Conjugation ready
Our carrier-free antibodies are typically supplied in a PBS-only formulation, purified and free of BSA, sodium azide and glycerol. This conjugation-ready format is designed for use with fluorochromes, metal isotopes, oligonucleotides, and enzymes, which makes them ideal for antibody labelling, functional and cell-based assays, flow-based assays (e.g. mass cytometry) and Multiplex Imaging applications.
Use our
conjugation kits
for antibody conjugates that are ready-to-use in as little as 20 minutes with 1 minute hands-on-time and 100% antibody recovery: available for fluorescent dyes, HRP, biotin and gold.
Sandwich ELISA
The recommended antibody orientation is based on internal optimization for ELISA-based assays. Antibody orientation is assay dependent and needs to be optimized for each assay type. Please note that the range provided for this antibody is only an estimation based on the performance of the product using the recommended antibody pair. Performance of the antibody pair will depend on the specific characteristics of your assay. We guarantee the product works in sandwich ELISA, but we do not guarantee the sensitivity or dynamic range of the antibody in your assay.

Orexin also known as hypocretin is a neuropeptide that regulates arousal wakefulness and appetite. It comprises two types orexin-A and orexin-B and has a mass of approximately 3.5 kDa. Orexin is primarily produced in the hypothalamus but its receptors are found widely across the brain particularly in areas involved in regulating sleep and feeding behaviors. Orexin neurons release neurotransmitters and project to multiple brain regions influencing various physiological processes.
Biological function summary
Orexin functions as an important player in maintaining wakefulness and energy homeostasis. It does not act alone but interacts with receptors forming part of complex neural networks. The orexin system stimulates neurons that release other excitatory neurotransmitters therefore enhancing wakefulness and alertness. Through its action on the autonomic nervous system orexin impacts metabolic rate and can drive food intake when energy levels are low.
Pathways
Orexin integrates into the sleep-wake regulatory network and energy balance pathways. It is involved in the sleep-wakefulness cycle by modulating the levels of neurotransmitters like noradrenaline and dopamine. Orexin interacts with the hypothalamic-pituitary-adrenal axis impacting stress responses and energy expenditure. The involvement in these pathways links it to other proteins like leptin and ghrelin which also participate in energy homeostasis.
Orexin has significant implications in the pathophysiology of narcolepsy and obesity. Narcolepsy often results from the loss of orexin-producing neurons leading to excessive daytime sleepiness and sudden muscle weakness. Orexin's involvement in energy regulation connects it to obesity where orexin deficiency or altered signaling may contribute to weight gain and metabolic issues. In the context of these disorders orexin's interactions with leptin become significant as leptin influences appetite and energy expenditure highlighting a connection within the regulatory network affecting both sleep and metabolism.
