The RhD protein also known as the Rhesus D antigen is a transmembrane protein that functions as a transporter across erythrocyte membranes. The protein has a mass of approximately 30 kDa. It is a part of the Rh blood group system and is most prominently expressed on the surface of red blood cells. The presence or absence of the RhD antigen on red blood cells defines the Rh positive or negative blood type which is important in transfusion medicine and pregnancy.
Biological function summary
The RhD protein is involved in transporting ammonium and carbon dioxide across the erythrocyte membrane. Although it is part of a larger Rh protein complex which consists of other Rh-associated glycoproteins RhD itself plays a critical role in maintaining the structural integrity and physiological function of red blood cells. The expression levels of RhD can affect the immunogenic response in situations like pregnancy where maternal and fetal blood types may differ.
Pathways
The RhD protein interacts with the RhCE protein to form complexes that participate in the Rh blood group system. This system is integral to the red blood cell membrane's structural organization and is linked to cellular processes that influence erythrocyte shape and deformability. The RhD protein also has interactions along pathways related to the anion exchanger protein AE1 impacting red cell ion transport and gas exchange.
Several conditions link to RhD notably hemolytic disease of the newborn and autoimmune hemolytic anemia. Hemolytic disease of the newborn occurs when an Rh-negative mother carries an Rh-positive fetus leading to maternal sensitization to the RhD antigen. This can result in maternal antibodies crossing the placenta and attacking fetal red blood cells. Autoimmune hemolytic anemia may also involve RhD if RhD-targeted antibodies are produced leading to destruction of red blood cells similar to the effect seen with other membrane proteins like the glycophorin C.
