SAV1 also known as Salvador homolog 1 is a scaffold protein that plays an important role in regulating cell proliferation and apoptosis. This protein has a molecular weight of around 45 kDa and is widely expressed in various tissues throughout the body including the lung liver and heart. SAV1 contains two WW domains which facilitate interactions with other proteins critical for its function in signaling pathways.
Biological function summary
SAV1 participates in the Hippo signaling pathway acting as a core component in the regulation of organ size by controlling cell growth and apoptosis. SAV1 forms a complex with other proteins such as MST1/2 kinases and LATS1/2 kinases promoting phosphorylation events that lead to the inhibition of the transcription co-activators YAP and TAZ. This inhibition prevents unchecked cell growth proving important for maintaining proper cellular homeostasis.
Pathways
SAV1 is essential in both the Hippo and MAPK signaling pathways. In the Hippo pathway it interacts closely with MST1/2 playing a role in pathway activation and signaling cascade propagation. In the MAPK pathway it indirectly influences cellular responses to external stress signals linking to proteins such as RAF1. The coordination within these pathways ensures regulated cell proliferation and stress response.
SAV1 is linked to cancer and cardiac hypertrophy. When the Hippo pathway malfunctions due to altered SAV1 expression it can lead to uncontrolled cell growth and tumorigenesis. In some cancer types like hepatocellular carcinoma SAV1 dysfunction correlates with aggressive tumor traits. Moreover its alteration affects cardiac hypertrophy through its interactions with proteins like YAP influencing heart muscle enlargement due to stress which can lead to heart failure.
