Complement Factor B also known as factor B complement is an important protein in the immune system. It weighs about 93 kDa and mainly expresses in the liver though it also circulates in the blood. This protein plays an important role in the alternative pathway of complement activation. Upon activation factor B combines with complement C3b to form the C3bB complex which then turns into the C3 convertase. This converts more C3 into C3b amplifying the immune response.
Biological function summary
Complement Factor B serves as a serine protease that is essential in the body's defense mechanisms. It is an integral part of the C3/C5 convertase enzyme complex. When C3b binds to factor B complement it splits into Ba and Bb fragments. The Bb fragment becomes part of the C3 convertase structure on microbial surfaces facilitating greater immune defense. Factor B's action allows the body to rapidly respond to pathogens contributing to innate immunity by helping opsonize and clear pathogens.
Pathways
This protein participates chiefly in the alternative complement pathway. It interacts closely with other complement components such as factor C3 and factor D to promote this pathway. The cascade initiated by complement factor B targets pathogens for destruction and assists in inflammation processes. It ensures a quick response in eliminating infectious agents before the adaptive immune system gets activated linking the innate and adaptive immune responses.
Research links Complement Factor B to conditions like age-related macular degeneration (AMD) and atypical hemolytic uremic syndrome (aHUS). In these disorders abnormal regulation or mutations in factor B can lead to excessive complement activation. For AMD this hyperactivity results in damage to retinal cells whereas in aHUS it causes issues in blood vessels and kidneys. Decay-accelerating factor and factor H also play notable roles here regulating this pathway to prevent overactivation that could harm healthy tissues.
