Calpain 5 also known as PalBH is a non-classical member of the calpain family with a mass of approximately 80 kilodaltons. Calpain 5 is made from a calcium-dependent cysteine protease showing expression in tissues like the retina and neurons. Known for its calpain-like protease domain and an extended C-terminal region Calpain 5 lacks the typical EF-hand motifs found in other calpains.
Biological function summary
Calpain 5 acts without forming complexes with calmodulin or calpastatin which is common in other calpain family members. It modulates cellular processes such as cytoskeleton remodeling and cellular signaling. It plays roles in cell cycle progression and apoptosis by regulating molecules involved in these processes. Calpain 5 interacts with cytoskeletal proteins and promotes degradation or remodeling facilitating normal cellular function and adaptation to stress.
Pathways
Calpain 5 functions are relevant in signal transduction and apoptosis pathways. In signal transduction it participates in protein kinase C-related pathways where it modulates protein activity critical for cell communication. Regarding apoptosis Calpain 5 interacts with BCL-2 family proteins which are key regulators of cell death influencing cell survival outcomes.
Mutations or dysregulation of Calpain 5 associate with retinal disorders like vitelliform macular dystrophy and cone-rod dystrophy. These conditions involve Calpain 5 in the deterioration of retinal cells likely through interactions with retinal proteins such as CRX and RDH12. Understanding Calpain 5's role in these disorders may offer insights into new therapeutic approaches for degeneration of retinal cells.
