Monkeypox virus A4L also known as DNA polymerase catalytic subunit plays a critical role in viral replication. This protein with an approximate mass of 110 kDa is expressed in the cytoplasm of infected host cells. A4L functions mechanistically as a DNA-dependent DNA polymerase facilitating the elongation of the DNA strand during replication. It achieves this by catalyzing the addition of deoxynucleotides to the 3'-end of the growing DNA chain using the viral DNA as a template.
Biological function summary
A4L is pivotal for the propagation of the monkeypox virus ensuring successful duplication of the viral genome. As an essential component of the viral replication complex it interacts with other viral and host proteins to execute its function. Its activity is indispensable for the replication cycle which directly influences the virus's ability to infect new cells and propagate throughout the host organism.
Pathways
Monkeypox virus A4L integrates into the virus replication and repair pathways. It interacts with replication proteins such as E9L DNA polymerase orchestrating a multifaceted machinery critical for viral DNA synthesis. A4L's role also aligns with recombination pathways where it participates in repairing damaged viral DNA ensuring the integrity and continuity of viral replication.
The activity of A4L is closely associated with the development and progression of monkeypox infection in humans. The protein's function is central to the pathogenicity of the virus implicating it in skin lesions and systemic involvement characteristic of the disease. The presence of related proteins like the host's replication enzymes might further modulate the severity and spread of infection making A4L a potential target for therapeutic intervention to control monkeypox outbreaks.
