SARS-CoV2 nsp4 or non-structural protein 4 is a component of the viral replication and transcription complex. Nsp4 is about 54 kilodaltons in mass and is produced by the cleavage of polyprotein 1a/1ab. This protein mainly expresses in host cells where the virus replicates. During infection nsp4 partners with other non-structural proteins to modulate the microenvironment needed for viral RNA synthesis forming part of the membrane-associated complex important for coronavirus replication.
Biological function summary
Nsp4 along with nsp3 and nsp6 plays an important role in remodeling host cell membranes to form double-membrane vesicles (DMVs) which serve as sites for viral replication. These vesicles protect viral RNA from host cell defenses. Nsp4 contributes to membrane rearrangements that create optimal conditions for viral replication complexes. By cooperating within this multi-protein machinery nsp4 facilitates efficient RNA synthesis and assembly of new virions.
