IgG4 also known as IgG4-Fc or IGHG4 is one of the subclasses of Immunoglobulin G (IgG) antibodies. Mechanically it plays a role in the immune response by recognizing and binding to specific antigens. Its molecular weight is approximately 146 kDa which is typical for IgG molecules. IgG4 is commonly expressed in the blood and extracellular fluids. It can also be found at sites of chronic inflammation reflecting its involvement in modulating immune responses.
Biological function summary
IgG4 antibodies act to regulate immune responses and prevent excessive inflammation. It does not engage in strong effector functions like other IgG subclasses due to its unique ability to exchange half-molecules leading to functional monovalency. This characteristic makes it part of an immune complex that can block other antibody functions by competitive binding without activating complement pathways. The IgG4 sequence shows unique properties that allow it to engage selectively in regulatory pathways which is a distinct attribute among immunoglobulins.

IgG4 relates to conditions like IgG4-related disease (IgG4-RD) and autoimmune diseases where it presents an abnormal increase in serum levels and tissue infiltration. IgG4-RD is characterized by elevated IgG4 protein and causes fibrotic tissue inflammation in organs such as the pancreas and salivary glands. IgG4's interaction with proteins like T regulatory cells may contribute to downregulating immune attacks in autoimmune disease highlighting its dual role in both defensive and regulatory immune activities.
