TRIM2 also known as tripartite motif-containing protein 2 is a protein that plays an essential role in protein ubiquitination. It has a molecular mass of approximately 75 kDa. TRIM2 is mainly found in neurons and has expression profiles in the brain and other neural tissues. The protein contains a RING-type zinc finger domain that facilitates its E3 ubiquitin ligase activity enabling it to tag specific proteins for degradation by the proteasome system.
Biological function summary
TRIM2 functions by modulating the stability of specific proteins involved in neuronal growth and maintenance. It operates as part of ubiquitination complexes allowing for targeted protein degradation. Neurodevelopmental processes rely on TRIM2 activity to maintain cellular homeostasis. Its involvement in axonal transport and stabilization of microtubule-associated proteins underlines its important role in nervous system integrity and functionality.
Pathways
TRIM2's function integrates into the ubiquitin-proteasome pathway and neural development processes. Within the ubiquitin-proteasome pathway TRIM2 interacts closely with other E3 ligases and substrates essential for proper cell functioning. It also relates to the Neurotrophin TRK receptor signaling pathway where TRIM2 can influence neuronal survival and differentiation. These pathways connect TRIM2 to proteins like TRK receptors and other ligases that manage protein turnover and neuronal signaling.
Disrupted TRIM2 function associates with neurological conditions such as Charcot-Marie-Tooth disease and neurodegenerative disorders. Mutations in TRIM2 lead to impaired ubiquitination activities contributing to these diseases' progression. TRIM2 dysfunction also links to abnormalities in proteins like neurofilament light chain which further aggravate disease pathology by destabilizing neuronal architectures and interrupting normal neural processes.
