Clonality:Monoclonal,
Clone number:DM1,
Isotype:IgG1,
Light chain type:kappa,
Carrier free:No,
Reacts with:Dengue virus 2,
Applications:WB, ELISA, ICC/IFSee reactivity dataSee the reactivity data table below for information on validated species and application combinations.,
Specificity:This antibody reacts with the pre-membrane glycoprotein of Dengue Virus 2 in Western blot. In ELISA, it recognises prM glycoprotein from Dengue Virus 1,2,3 and 4.Using non-reduced and unheated samples to preserve conformational epitopes, as this antibodies recognize conformational rather than linear epitopes.

The Dengue Virus prM glycoprotein also known as pre-membrane protein is a critical component in the lifecycle of the dengue virus. This glycoprotein has a molecular mass of approximately 22 to 23 kDa. It is expressed on the surface of immature virions in host cells where it undergoes conformational changes. During viral maturation prM shields the fusion loop of the envelope (E) protein preventing premature fusion during viral replication and assembly. This action supports the assembly of mature infectious virions by eventually cleaving into the membrane (M) protein under the influence of furin protease.
Biological function summary
The Dengue Virus prM glycoprotein plays an important role in virus maturation and infectivity. It forms a complex with the envelope glycoprotein to protect viral components during transport and to ensure proper assembly. This complex is necessary for the virus to remain intact as it exits the host cell facilitating successful infection of new cells. The cleavage of prM into the M protein by cellular proteases remains an indispensable process that dictates the infectiosity and virulence of the virus.
Pathways
The role of the prM protein is highly significant in the viral entry and maturation pathways. The cleavage of prM glycoprotein occurs in the endoplasmic reticulum and trans-Golgi network involving key cellular processes that modify viral particles. This process is closely associated with the functionality of other viral proteins such as the envelope (E) protein which contributes to the virus’s ability to bind and fuse with host cell membranes. This coordination ensures the efficient production of mature dengue virions ready for replication.
The Dengue Virus prM glycoprotein is a major focus in the study of dengue fever and severe dengue like dengue hemorrhagic fever. The prM protein's involvement in the maturation pathway directly affects virulence and infectivity making it an attractive target for vaccine development and therapeutic interventions. During infection antibodies developed against prM and the related envelope (E) protein can aid in neutralization but they may also contribute to antibody-dependent enhancement (ADE) which can exacerbate the severity of the disease. Understanding prM's role provides insight into potential strategies for preventing and managing dengue-related conditions.
