ELISA (Enzyme-Linked Immunosorbent Assay) kit (ab46074) is designed for the quantitative measurement of TRAIL in cell culture supernatants, buffered solutions or Human serum, plasma, and other body fluids.
A monoclonal antibody specific for TRAIL has been coated onto the wells of the microtiter strips provided. Samples, including standards of known TRAIL concentrations, control specimens or unknowns are pipetted into these wells. During the first incubation, the standards or samples and a biotinylated monoclonal antibody specific for TRAIL are simultaneously incubated. After washing, the enzyme Streptavidin-HRP, that binds the biotinylated antibody is added, incubated and washed. A TMB substrate solution is added which acts on the bound enzyme to induce a colored reaction product. The intensity of this colored product is directly proportional to the concentration of TRAIL present in the samples.
This kit will recognize both endogenous and recombinant Human TRAIL.

TRAIL also known as TNF-related apoptosis-inducing ligand plays an important role in inducing apoptosis. This protein has a mass of around 32 kDa and is expressed across various tissues including immune cells. TRAIL interacts with specific receptors on the cell surface to initiate a signaling cascade that results in programmed cell death. It is an attractive therapeutic target because of its selective ability to induce apoptosis in cancer cells while sparing most normal cells.
Biological function summary
TRAIL's function extends beyond inducing cell death. It is an important member of the tumor necrosis factor (TNF) superfamily which regulates immune responses and inflammation. TRAIL has a significant connection with cancer and autoimmune diseases. TRAIL plays a dual role in cancer where it acts as a tumor suppressor but can also promote tumor progression if cancer cells become resistant to its effects. In cancer TRAIL’s interaction with proteins like FLIP can alter the apoptotic potential of the cells. In autoimmune diseases TRAIL's dysregulation can contribute to tissue damage due to increased immune activity highlighting its complex role in maintaining immune balance.
