Bradykinin also known as bradykinin (BK) is a peptide that plays an important role in multiple physiological processes. Bradykinin consists of nine amino acids and typically has a molecular weight of around 1060 daltons. It is primarily expressed in endothelial cells and can be produced by blood plasma proteins in response to specific stimuli. Bradykinin exemplifies a mediator of inflammation and vasodilation contributing significantly to vascular permeability and lowering blood pressure.
Biological function summary
This peptide acts as an important modulator in inflammatory and pain processes. It forms part of the kallikrein-kinin system where it interacts with specific G-protein-coupled receptors known as bradykinin receptor B1 and B2. Bradykinin contributes to complex biological functions including the dilation of blood vessels smooth muscle contraction and activation of pain fiber nerve endings. This activity makes it critical for the body’s response to inflammation and injury.
Pathways
Bradykinin participates in the kallikrein-kinin system and the renin-angiotensin system. In the renin-angiotensin system bradykinin is related to angiotensin-converting enzyme (ACE) which degrades bradykinin and counters its vasodilatory effects. Bradykinin's role in inflammation and blood pressure regulation is linked closely to other vasoactive peptides and cytokines contributing to its importance in diverse physiological pathways.
Bradykinin is associated with conditions such as hereditary angioedema and chronic cough. In hereditary angioedema excessive bradykinin production leads to recurrent episodes of swelling due to its capacity to increase vascular permeability. The peptide also plays a role in inducing a chronic cough as an adverse reaction via its action on sensory neurons in the respiratory tract. This is often observed in patients using ACE inhibitors as the inhibition leads to increased levels of bradykinin.
