Poliovirus type 3 also known as PV3 is a strain of the poliovirus responsible for causing poliomyelitis. It is a member of the Picornaviridae family and is an RNA virus with a single-stranded positive-sense genome approximately 7500 nucleotides in length. The viral capsid proteins including VP1 VP2 VP3 and VP4 assemble to form the icosahedral structure with a mass of about 8000 kDa. This structure facilitates the viral entry into host cells through poliovirus receptor CD155 which is expressed on a variety of human cells particularly epithelial cells in the intestinal tract and motor neurons.
Biological function summary
The replication process of poliovirus type 3 involves cellular mechanisms where the virus hijacks the host's ribosomes for protein synthesis. The viral RNA is directly translated into a polyprotein that is subsequently cleaved by viral proteases into structural and non-structural proteins necessary for viral assembly and replication. These processes take place in the cytoplasm and involve the use of host cellular machinery including membrane vesicle formation. Poliovirus type 3 does not exist as part of a stable complex but interacts transiently with cellular components throughout its lifecycle.

Poliovirus type 3 disrupts host cell functions particularly those involved in cellular protein synthesis and apoptosis. It interacts with signaling pathways such as the PI3K/Akt pathway significantly affecting cell survival and death. The virus-induced cleavage of host proteins like eIF4G and the alteration of the host translation machinery are critical for viral propagation and are actions closely linked with these pathways. These interactions enable the virus to evade host immune responses while maximizing its replication.
Poliovirus type 3 is directly linked to poliomyelitis which can lead to irreversible paralysis. The infection involves the invasion and destruction of motor neurons linked to proteins such as caspases activated during cell apoptosis induced by the virus. The disruption of muscular function and neural communication due to motor neuron damage is an important characteristic of poliovirus type 3 pathogenesis. Additionally while less common the virus is associated with post-polio syndrome where affected individuals experience muscle weakness years after recovery from poliomyelitis.
