SEMA3B also known as Semaphorin-3B is a member of the semaphorin protein family functioning as a guidance cue in neural development. It has an approximate mass of 85 kDa. SEMA3B expresses in various tissues including the nervous system lungs and kidneys. The protein interacts with neuropilin and plexin receptors to mediate its mechanical effects in cell signaling and axonal guidance.
Biological function summary
SEMA3B plays a role in modulating cell migration and apoptosis. As part of the semaphorin family it does not form complexes but signals through receptor interactions. In the nervous system SEMA3B contributes to neural network formation by guiding developing neurons. In addition to its neuronal functions SEMA3B participates in controlling angiogenesis and epithelial cell proliferation acting as a tumor suppressor.
Pathways
SEMA3B is involved in the axon guidance and PI3K/AKT signaling pathways. These pathways are important for cellular movement and survival. SEMA3B interacts with proteins such as neuropilin-1 (NRP1) and plexin-A (PLXNA) which are integral to its signaling pathway. Through these interactions SEMA3B regulates cellular behavior linking it to both neural development and cancer biology.
SEMA3B has strong links to cancer particularly lung and breast cancer where it functions as a tumor suppressor. Altered expression of SEMA3B is observed in these conditions leading to disrupted cellular signaling and uncontrolled cell proliferation. Additionally changes in SEMA3B expression levels associate with neuropilin-1 and other receptors influencing cancer progression and metastasis.
