Adiponectin receptor 1 (ADIPOR1) is a protein that mediates the effects of adiponectin a hormone involved in glucose regulation and fatty acid breakdown. ADIPOR1 is also known as the adiponectin receptor protein 1. It has a molecular weight of approximately 42 kDa. This receptor is widely expressed in various tissues including skeletal muscle liver pancreas and adipose tissue demonstrating its broad role in metabolism.
Biological function summary
ADIPOR1 facilitates the metabolic actions of adiponectin by increasing insulin sensitivity and promoting fatty acid oxidation. The receptor does not function alone; it operates in coordination with another isoform adiponectin receptor 2 (ADIPOR2) forming a complex to exert its full biological effects. This dual receptor system supports the regulation of energy homeostasis and inflammation influencing metabolic rate and improving lipid profiles.
Pathways
ADIPOR1 plays a critical role in the AMP-activated protein kinase (AMPK) pathway and the peroxisome proliferator-activated receptor alpha (PPAR-α) signaling pathway. These pathways are important for cellular energy balance and lipid metabolism. AMPK activation by ADIPOR1 leads to enhanced glucose uptake and fatty acid oxidation while interaction within the PPAR-α pathway regulates gene expression related to lipid metabolism. The activity of ADIPOR1 is intricately tied to other metabolic proteins like leptin and insulin thereby linking with these pathways.
ADIPOR1 involvement is significant in the development of type 2 diabetes and non-alcoholic fatty liver disease (NAFLD). Its interaction with adiponectin and the modulation of insulin sensitivity positions it central to the pathophysiology of these conditions. These interactions highlight its potential as a therapeutic target. Disruption in ADIPOR1 function may influence related proteins such as adiponectin and insulin contributing to the progression of metabolic disorders.
