RFXAP also known as Regulatory Factor X Associated Protein plays a role as a component in the transcriptional activation of MHC class II genes. This protein modulates gene expression by forming a complex with other transcription factors to bind to specific sequences in the MHC class II promoter region. It has a molecular mass of approximately 31 kDa. RFXAP is ubiquitously expressed in various tissues demonstrating a broad pattern that supports its fundamental function in cellular processes involved with immune response.
Biological function summary
RFXAP functions as an integral part of the RFX complex which also includes RFX5 and RFXANK. This complex acts as a transcriptional activator for MHC class II genes necessary for antigen presentation to CD4+ T lymphocytes. By participating in the assembly of the enhanceosome on the SXY module of MHC class II promoters RFXAP ensures regulated and proper transcription of these genes affecting immune system efficiency and adaptability.
Pathways
RFXAP contributes directly to the MHC class II expression pathway essential for adaptive immunity. It interacts and works closely with RFX5 and RFXANK within the same regulatory complex facilitating the transcription of major histocompatibility complex class II molecules. Proper function of these molecules is necessary for the recognition and processing of foreign antigens affecting how the immune system responds to pathogens.
Mutations or functional disruptions of RFXAP are linked to Bare Lymphocyte Syndrome Type II a condition characterized by the absence of MHC class II expression on antigen-presenting cells leading to severe immunodeficiency. In this disorder the connection with other proteins like RFX5 and RFXANK is critical since defects in any of these components can result in similar pathological outcomes highlighting the importance of RFXAP within the MHC class II transcriptional machinery.
