E2F6 a member of the E2F family of transcription factors acts as a transcriptional repressor. It regulates genes essential for cell cycle progression particularly those involved in DNA replication. E2F6 lacks the transactivation domain present in other E2F family members allowing it uniquely to repress transcription without activating it. E2F6 has a molecular mass of approximately 30 kDa. The protein is mostly expressed in the nucleus of proliferating cells but it is also found in certain other tissues including the brain and testes.
Biological function summary
E2F6 plays an important role in regulating the cell cycle. It belongs to a multiprotein complex called the repressive E2F6 complex or PRC1 (Polycomb Repressive Complex 1). This complex is responsible for chromatin remodeling and transcriptional repression of target genes important in S phase entry and mitosis. By repressing specific E2F-responsive genes E2F6 inhibits excessive cell proliferation therefore maintaining proper cell cycle control.
Pathways
E2F6 contributes to important pathways like the cell cycle and DNA damage response pathways. In these pathways it is involved in controlling the transition phases of the cell cycle particularly regulating entry into the S phase. E2F6 interacts with proteins such as Rb (retinoblastoma protein) and other E2F members like E2F1 and E2F4 to control cell growth and ensure genome integrity.
Dysregulation of E2F6 has links to cancer and developmental disorders. In cancer overexpression or mutations affecting E2F6 can lead to uncontrolled cell proliferation due to improper repression of cell cycle genes. Additionally defects in the E2F6 gene have associations with developmental syndromes through interactions with other proteins like EZH2 a component of the Polycomb group proteins. These associations highlight the importance of E2F6 in both normal cell function and disease states.
