RXRG also known as retinoid X receptor gamma is a nuclear receptor protein weighing approximately 55 kDa. It binds retinoic acids and mediates their effects in the cell. RXRG can form homodimers or heterodimers with other nuclear receptors. The protein's expression occurs mainly in the brain muscles and the spleen. As a transcription factor it regulates gene expression by binding to specific DNA sequences influencing various cellular processes.
Biological function summary
RXRG acts as a transcriptional regulator in the retinoid signaling pathway. It forms part of a heterodimer complex with receptors such as PPARs (peroxisome proliferator-activated receptors) and RXR α and β subunits. These complexes control genes involved in cell differentiation proliferation and apoptosis. Through RXRG cells can transduce external retinoid signals into specific gene expression responses impacting development and metabolic processes.
Pathways
RXRG integrates into significant signaling cascades including the retinoic acid signaling pathway and the adipogenesis pathway. It often partners with other nuclear receptors like RARs (retinoic acid receptors) and PPARs to mediate these pathways. For instance in the adipogenesis pathway RXRG's interaction with PPARγ regulates adipocyte differentiation and lipid metabolism. These pathways link to extensive roles in development and metabolic regulation.
RXRG plays a role in conditions such as obesity and certain types of cancer. In obesity the interaction between RXRG and PPARγ influences fat cell formation and energy balance. Additionally dysregulation of RXRG expression associates with cancers where it interacts with proteins like RXRα to alter cell proliferation and apoptosis. Understanding these associations highlights RXRG's importance in therapeutic research and potential drug targets.
