TBC1D1 also known as TBC1 domain family member 1 is a protein involved in the regulation of glucose metabolism. It acts as a GTPase-activating protein (GAP) specifically for the Rab family of small GTPases. TBC1D1 plays a role in the activation and translocation of glucose transporter type 4 (GLUT4) to the plasma membrane. This protein has a molecular mass of approximately 130 kDa. It is highly expressed in skeletal muscle and brown adipose tissue indicating its role in energy homeostasis.
Biological function summary
TBC1D1 participates in the regulation of insulin-stimulated glucose uptake. It is not part of a multi-protein complex but it interacts with other proteins to execute its functions. TBC1D1 has a significant role in metabolic processes influencing how cells respond to insulin. By acting on GLUT4 TBC1D1 facilitates glucose entry into cells which is an important step in maintaining glucose levels in the blood.
Pathways
TBC1D1 is critical in the insulin signaling and AMP-activated protein kinase (AMPK) pathways. It interacts with proteins such as Akt and AS160 which are important in mediating the effects of insulin. In the insulin signaling pathway TBC1D1 regulates GLUT4 translocation while in the AMPK pathway it contributes to energy balance and metabolism. These pathways highlight the role of TBC1D1 in maintaining cellular energy efficiency and glucose homeostasis.
TBC1D1 has relevance to obesity and type 2 diabetes. Variants in the TBC1D1 gene have been associated with obesity suggesting the gene's role in body weight regulation. Additionally disruptions in TBC1D1 function might influence insulin resistance an important feature of type 2 diabetes. The protein plays a part in this by regulating GLUT4 translocation and glucose uptake associating it with metabolic syndrome components.
