RAB43 also known as Ras-related protein Rab-43 is a member of the Ras superfamily of small GTPases specific for regulating intracellular vesicle trafficking. It has a molecular mass of about 25 kDa. RAB43 expresses in a variety of tissues with significant presence in the Golgi apparatus and endosomal compartments. These locations suggest its involvement in processes related to membrane trafficking and vesicular transport.
Biological function summary
The small GTPase RAB43 plays an important role in intracellular membrane trafficking and cargo sorting. It participates in a multi-protein complex that includes other small GTPases that help regulate membrane fusion events. RAB43 ensures accurate transport of proteins and lipids between the Golgi apparatus and various endosomal subcompartments. This regulation facilitates proper cellular function and maintenance of cellular architecture.
Pathways
Molecular studies have shown that RAB43 influences pathways such as endocytic trafficking and protein processing. It interacts with proteins involved in the vesicular transport pathway ensuring efficient transport of certain cargo molecules to their respective destinations. Notably RAB43 associates with proteins such as SNAREs within these pathways indicating its role in docking and fusion of transport vesicles.
Recent research highlights RAB43's involvement in neurodegenerative conditions such as Parkinson's disease and certain developmental disorders. Alterations in RAB43 expression or function disrupt vesicle trafficking potentially leading to neuronal damage. The interplay between RAB43 and proteins like LRRK2 which have significant links to Parkinson's disease supports its relevance in neurological health and disease.
