VEGF 121B also known as Vascular Endothelial Growth Factor 121B is an isoform of the VEGF family. Mechanically it functions as a signal protein that stimulates vasculogenesis and angiogenesis. VEGF 121B is a splice variant that lacks exon 6 and 7 and has a mass of approximately 14 kDa. This protein is expressed in a variety of tissues but it is reported mainly in endothelial cells where it promotes the growth of blood vessels.
Biological function summary
Vascular Endothelial Growth Factor 121B plays an important role in the formation of new blood vessels. It functions as part of the larger VEGF protein family complex which includes other members such as VEGF165 and VEGF189. The binding of VEGF 121B to its receptors on endothelial cells activates signaling pathways that regulate vascular development and permeability. Its role extends to facilitating tissue repair and regeneration by enhancing the supply of nutrients and oxygen through blood vessel growth.
Pathways
VEGF 121B actively participates in the angiogenesis and vascular endothelial growth factor receptor (VEGFR) signaling pathways. These pathways involve interactions with proteins such as VEGFR1 and VEGFR2 which mediate its effects on endothelial cells. VEGF 121B binds to these receptors and triggers downstream signaling cascades ultimately promoting cell proliferation migration and formation of vascular networks.
Altered expression or dysfunctional signaling of VEGF 121B relates to cancer and age-related macular degeneration (AMD). In cancer overexpression of VEGF 121B can lead to increased tumor vascularization enabling tumor growth and metastasis. In AMD its imbalance could contribute to abnormal blood vessel growth in the retina. The dysregulation of VEGF 121B in these conditions often associates with other proteins in the VEGF family such as VEGF-A which together influence the disease progression and response to treatment.
