Alpha A Crystallin and Alpha B Crystallin are small heat shock proteins with molecular masses of approximately 20 kDa each. These proteins also known as HspB4 and HspB5 respectively serve as molecular chaperones. They prevent the aggregation of other proteins under stress conditions. Both crystallins express abundantly in the lens of the eye ensuring eye transparency. While Alpha A Crystallin mainly localizes in the lens Alpha B Crystallin appears more ubiquitously found in the eye heart skeletal muscle kidney and brain.
Biological function summary
The two crystallins play essential roles in maintaining cellular homeostasis and protein integrity. They often form large oligomeric complexes enabling them to sequester unfolded proteins aiding in proper folding. Alpha B Crystallin in particular acts as a stress-resilient protein helping cells survive under conditions like heat oxidative stress and osmotic pressure. These functional properties link the crystallins to protective roles in cells guarding against permanent damage from misfolded proteins.
Pathways
Alpha A and Alpha B Crystallins both integrate into the proteostasis network particularly in pathways related to protein folding and stress responses. They interact closely with proteins such as Hsp70 which assists in the ATP-dependent folding of proteins. Both crystallins also play roles in the MAPK/ERK pathway which regulates cell survival and proliferation. This involvement underlines their function in managing cell stress and promoting recovery and repair processes.
These crystallins have associations with cataracts and neurodegenerative diseases. Alpha A Crystallin mutations often link to hereditary cataracts while Alpha B Crystallin's role extends to neurodegenerative disorders like Alzheimer's disease. In Alzheimer's it interacts with Tau protein contributing to the pathological process. The involvement of these crystallins in various diseases highlights the importance of their normal function in cellular protection and stress response mechanisms.
