SLITRK4 also known as SLIT and NTRK-like protein 4 is a protein involved in neural cell signaling. It is a member of the SLITRK family which consists of six known proteins that share structural similarities to both the Slit and Trk families of proteins. SLITRK4 has a molecular mass of approximately 104 kDa. This protein is highly expressed in the brain particularly in the thalamus region as well as in some other neural tissues. Its expression is less present in peripheral tissues indicating a more central nervous system-specific role.
Biological function summary
SLITRK4 plays an essential role in modulating synaptic activity and neuron-neuron communication. It is part of a transmembrane protein family interacting with other synaptic and intracellular proteins to coordinate signaling events that influence neuronal growth and connectivity. By regulating synaptic plasticity SLITRK4 contributes to the formation of functional neural circuits and is involved in the proper development and maintenance of synapses.
Pathways
SLITRK4 is integrally engaged in neurodevelopmental pathways particularly those governing neural circuit assembly and synaptic function. It interacts with proteins like leucine-rich repeat transmembrane neuronal proteins to facilitate synaptic adhesion and signaling processes. Additionally SLITRK4's role in synaptic modulation links it to pathways controlling neurotransmitter release and receptor signaling essential for effective communication between neurons.
SLITRK4 has shown associations with neurodevelopmental disorders such as schizophrenia and autism spectrum disorders. Disruptions in SLITRK4 can lead to misregulated synaptic function contributing to the pathophysiology of these conditions. Alterations in its expression or function may also affect interactions with related proteins like neurexins which are important for synaptic stability and network formation. Understanding these connections continues to provide insights into the molecular bases of these complex disorders.
