NDFIP2 also known as NEDD4 family-interacting protein 2 is an E3 ubiquitin ligase adaptor protein that plays a role in protein ubiquitination and degradation. It has a mass of approximately 24 kDa. NDFIP2 is expressed in various tissues including high levels in the brain and immune cells. It localizes to the cytoplasm and interacts with several pathways involving protein trafficking and cellular stress responses.
Biological function summary
NDFIP2 interacts with NEDD4 family E3 ubiquitin-protein ligases and plays a role in inhibiting cell surface receptor signaling by mediating the ubiquitination and subsequent degradation of the receptor complex. It participates in the regulation of adaptive immune responses and its activity adjusts according to cellular stress signals adapting the protein machinery to ensure cellular function. NDFIP2 does not form part of any complex but directly interacts with enzymes that tag proteins for degradation.
Pathways
NDFIP2 influences ubiquitin-mediated protein degradation and the JAK-STAT signaling pathway. In the ubiquitin pathway NDFIP2 works closely with NEDD4 facilitating tagging of proteins for proteasome-mediated destruction. In the JAK-STAT pathway its role in receptor regulation indirectly influences signal transduction and the expression of genes vital for immune function and growth regulation.
NDFIP2 is linked with conditions like neurodegenerative disorders and autoimmune diseases. Its expression and function alterations can influence disease outcomes by disrupting protein degradation and receptor signaling. For instance NDFIP2 interacts with proteins like NEDD4 and JAK which are involved in pathways implicated in the progression of specific neurodegenerative disorders as well as in autoimmune conditions where dysfunctional signaling leads to improper immune response.
