ABHD12 also known as alpha/beta-hydrolase domain-containing protein 12 is an enzyme that mechanically functions as a lipase. It plays a role in catalytic activity by hydrolyzing lysophospholipids and other lipid substrates. ABHD12 is a membrane-bound protein with a molecular mass of approximately 45 kDa. It is widely expressed in the human body with higher expressions noted in the brain immune system and liver.
Biological function summary
ABHD12 plays an important role in the immune and central nervous systems. It is not currently identified as part of a complex but is significant in regulating lipid signaling and homeostasis. By maintaining proper lysophosphatidylserine and lysophosphatidic acid levels ABHD12 influences neuroinflammation and neurodegenerative processes.
Pathways
ABHD12 interacts with lipid signaling and metabolic pathways. It is involved in the endocannabinoid signaling pathway and the lysophospholipid signaling pathway. Within these pathways important connections exist between ABHD12 and other proteins like MAGL (monoacylglycerol lipase) and FAAH (fatty acid amide hydrolase) which help modulate the levels of lipid messengers.
ABHD12 has connections to neurodegenerative diseases including PHARC syndrome and multiple sclerosis. Mutations in the ABHD12 gene result in PHARC syndrome which involves demyelination and peripheral neuropathy. In the context of multiple sclerosis ABHD12’s role in neuroinflammation relates it to proteins such as TNF-alpha and IL-1 beta due to their involvement in immune responses and inflammatory processes.
