ST6GalNAc I also known as ST6 N-acetylgalactosaminide alpha-26-sialyltransferase 1 plays an important mechanical role in glycosylation processes. It adds sialic acid to the terminal position of N-acetylgalactosamine on glycoproteins. The enzyme has a molecular weight of around 55 kDa. Expression of ST6GalNAc I is found predominantly in the Golgi apparatus where glycosylation occurs. It is expressed in tissues such as the liver heart and brain though expression levels can vary.
Biological function summary
ST6GalNAc I is involved in the regulation of cell-to-cell interactions and immune responses. It is not known to be a part of larger enzyme complexes but operates independently adding sialic acid residues that can modify the properties and functions of glycoproteins on the cell surface. These modifications can influence cell signaling migration and adhesion through their effects on the cell glycocalyx.

ST6GalNAc I plays a role in the sialylation pathway which affects processes like cellular communication and protein stability. The enzyme's activity influences the O-glycan biosynthesis pathway. It works alongside other proteins such as ST3Gal I and ST6Gal I to fine-tune the sialylation states of glycoconjugates. ST6GalNAc I has a link to cancer and autoimmune disorders. Abnormal expression or activity of ST6GalNAc I can alter glycosylation patterns contributing to the progression of colon cancer by influencing tumor cell metastasis and immune cell evasion.
