KLF10 also known as TIEG1 is a transcription factor belonging to the Kruppel-like factor family. It weighs approximately 54 kDa and is expressed in various tissues including bone liver and muscle. KLF10 plays an important role in regulating gene expression influencing numerous cellular processes such as proliferation differentiation and apoptosis. Its expression levels often respond to external stimuli like transforming growth factor-beta (TGF-β) reflecting its versatile function in cellular responses.
Biological function summary
KLF10 affects several pathways through its interactions with other proteins and DNA elements. As a member of the human transcription machinery it acts as a coregulator often partnering with SMAD proteins to modulate gene expression in TGF-β signaling. KLF10 influences cellular activities by regulating genes involved in tissue development and repair making it integral to maintaining cellular homeostasis and function across numerous organ systems.
Pathways
This protein integrates into key signaling cascades most notably TGF-β and bone morphogenetic protein (BMP) signaling pathways. In TGF-β signaling KLF10 works closely with SMAD4 to regulate genes that control cellular growth and differentiation. This partnership is important for transmitting signals that govern cell fate decisions often influencing how cells respond to their immediate environment and driving critical cellular processes.
KLF10 links to conditions such as osteoporosis and cancer. Alterations in its expression or function can disrupt normal cellular processes contributing to the pathogenesis of these diseases. For example in osteoporosis KLF10's role in bone remodeling and maintenance becomes dysregulated affecting bone density and strength. In cancer its interactions with other proteins like SMAD4 and p53 can influence cell cycle control and tumor suppression highlighting its dual role in both health and disease progression.
