beta Hydroxybutyrate (beta HB) Assay Kit (Colorimetric) (ab83390) utilizes beta HB Dehydrogenase to generate a product which reacts with a colorimetric probe with an absorbance band at 450 nm.
The kit is an easy and accurate assay to measure beta HB levels in biological samples. The assay is linear for 1-20 nmol beta HB in up to 100 μL samples or 0.01-0.2 mM of beta HB samples.

This product is manufactured by BioVision, an Abcam company and was previously called K632 β-Hydroxybutyrate (β-HB) Colorimetric Assay Kit. K632-100 is the same size as the 100 test size of ab83390.
During diabetic ketoacidosis the liver produces ketone bodies (acetoacetate, acetone, and primarily beta-hydroxybutyrate) from the acetyl CoA produced from fatty acid oxidation. The very high rate of ketone body production outstrips the body's ability to utilize them as an energy source and the blood concentration builds up. As rather strong acids, they result in a significant drop in blood pH.

Beta-Hydroxybutyrate also called β-hydroxybutyrate or B-hydroxybutyrate serves as an important ketone body in the metabolism of energy. It has a molecular mass of approximately 104.1 g/mol. Cells in the liver particularly during periods of low glucose availability produce beta-hydroxybutyrate from acetyl-CoA derived from fatty acid oxidation. The liver then releases it into the bloodstream where peripheral tissues such as the brain heart and skeletal muscle can utilize it as an energy source. Researchers commonly use beta-hydroxybutyrate assays to quantify its levels in serum or plasma providing valuable information about metabolic status and energy balance.
Biological function summary
Beta-hydroxybutyrate functions as both a fuel molecule and a signaling molecule. It is part of the ketone body complex which also includes acetoacetate and acetone. It assists in providing energy to tissues during fasting or ketogenic diets by entering cells and converting back to acetyl-CoA which enters the citric acid cycle. Additionally beta-hydroxybutyrate influences gene expression inflammation and oxidative stress by interacting with receptors and modulating pathways at the cellular level.
Pathways
Beta-hydroxybutyrate is involved in the ketogenesis and ketolysis pathways. During ketogenesis liver cells convert fatty acids to ketone bodies including beta-hydroxybutyrate which is then transported to extrahepatic tissues. In these tissues ketolysis occurs refashioning the ketone body back to acetyl-CoA. The enzyme 3-hydroxybutyrate dehydrogenase plays an important role in this process by interconverting beta-hydroxybutyrate and acetoacetate. These pathways are closely linked to gluconeogenesis and glycolysis underlining the significance of beta-hydroxybutyrate in energy homeostasis.
Beta-hydroxybutyrate levels are highly relevant in conditions like diabetes and epilepsy. In diabetes particularly in type 1 diabetic ketoacidosis elevated beta-hydroxybutyrate levels result from uncontrolled ketogenesis due to insufficient insulin highlighting metabolic dysregulation. Meanwhile in epilepsy ketogenic diets aim to stabilize neuronal activity by increasing beta-hydroxybutyrate levels which has been observed to reduce seizure frequency. Proteins such as insulin in diabetes and glutamate transporters in epilepsy play roles in these contexts showing the diverse impact of beta-hydroxybutyrate on health and disease.
