The Notch1 intracellular domain often called NICD plays a significant role in cell signaling. NICD is approximately 120 kDa in mass and is primarily expressed in various tissues especially during development. It also shows expression in multiple cell types including T-cells in the immune system. The Notch1 protein undergoes proteolytic cleavage to release the NICD which then translocates to the nucleus to influence gene expression.

The Notch1 intracellular domain acts as a transcriptional regulator within the Notch signaling pathway. It forms a complex with other proteins including the transcription factors RBPJ (Recombination Signal Binding Protein for Immunoglobulin Kappa J Region) and MAML (Mastermind-like) to modulate the transcription of target genes.
