Tau412 also known as Tau is a microtubule-associated protein that plays a role in stabilizing microtubules. It has a molecular weight of approximately 55-62 kDa depending on its isoform. Tau is abundantly expressed in neurons but can also be found in other tissues like the heart and kidneys. Its mechanical function involves interaction with tubulin which is important for maintaining the structure of axons. Tau has six isoforms as a result of alternative splicing from a single gene located on chromosome 17.
Biological function summary
Tau412 helps in the assembly and stabilization of microtubules which are important for axonal transport and neuronal function. It is not part of a larger protein complex but interacts with several microtubule and cytoskeleton proteins. Tau phosphorylation regulates its binding with microtubules affecting microtubule stability. Imbalance in Tau phosphorylation states can disrupt cellular processes and result in aggregation.
Pathways
Tau412 is involved in the microtubule disassembly and assembly pathways. It interacts with proteins like glycogen synthase kinase-3 beta (GSK-3β) and CDK5 which phosphorylate Tau and regulate its interaction with microtubules. Through these pathways Tau affects cellular transport and signaling mechanisms essential for neuronal function and survival.
Mutations or misregulation of Tau412 are linked to neurodegenerative diseases such as Alzheimer's disease and frontotemporal dementia. In Alzheimer's disease hyperphosphorylated Tau forms neurofibrillary tangles which are hallmarks of the disease along with amyloid-beta plaques. Tauopathies a group of disorders characterized by accumulations of Tau often involve interactions with amyloid-beta and other proteins such as apolipoprotein E (ApoE) which influence disease pathology and progression.
