ELK4 also known as SAP-1 (Serum Response Factor Accessory Protein 1) functions as a transcription factor within the Ets family. ELK4's molecular mass is approximately 51 kDa. This protein is expressed widely in various tissues including the brain heart and lungs. It binds to the DNA sequence GGAA/T to regulate gene expression and plays a significant role in cellular processes by modulating transcriptional response to external stimuli.
Biological function summary
The ELK4 protein influences cell proliferation and differentiation. It forms a complex with serum response factor (SRF) to modulate the activity of immediate early genes like c-fos. The ELK4-SRF complex binds to serum response elements (SREs) within promoter regions of target genes thereby impacting cellular responses such as growth and survival. Additionally ELK4's interactions with other proteins in the nucleus enhance its function in regulating gene expression.
Pathways
The ELK4 protein plays an important role in the MAPK/ERK signaling pathway which is important for transmitting extracellular signals to the nucleus and affecting gene expression. It interacts closely with other proteins such as ERK1/2 leading to its phosphorylation and activation. Also ELK4 participates in the Ras signaling cascade further influencing cellular responses to growth factors.
ELK4's dysfunctional regulation contributes to cancer particularly in prostate carcinoma. Aberrant ELK4 signaling affects the expression of genes related to cell proliferation and apoptosis supporting tumorigenesis. Additionally ELK4 associates with the SRF protein in the development of cardiovascular disorders where anomalous signaling can lead to pathological cardiac hypertrophy and failure.
