Centromere protein T commonly known as CENPT is an important component of the kinetochore complex. This protein has a molecular mass of approximately 94 kDa and plays an essential role in chromosome segregation during cell division. CENPT primarily localizes to the centromere region of chromosomes and is ubiquitously expressed in dividing cells particularly in human tissues with high cellular turnover. Often referred to as CENP-T it several times forms part of the inner kinetochore binding and directing microtubules for accurate chromosomal movement.
Biological function summary
Centromere protein T engages in stabilizing kinetochores ensuring proper chromosome alignment and segregation. As a part of the constitutive centromere-associated network (CCAN) complex CENPT cooperates closely with other proteins such as CENP-C and CENP-A to maintain kinetochore structure and function. This protein actively participates in facilitating the attachment of spindle microtubules to the centromere orchestrating the dynamic interactions necessary for efficient cell division.
Pathways
The role of CENPT integrates into the mechanistic regulation of the mitotic cell cycle. CENPT is pivotal in the pathway driven by the anaphase-promoting complex/cyclosome (APC/C) which regulates the separation of sister chromatids. Moreover CENPT works in conjunction with the spindle assembly checkpoint ensuring that chromosomes are properly aligned before anaphase onset. This coordination involves interactions with proteins like BUB1 and MAD2 safeguarding the fidelity of chromosome transmission during mitosis.
Centromere protein T has been implicated in chromosomal instability which is a hallmark of certain cancers. Abnormal expression or function of CENPT can lead to aneuploidy contributing to the progression of conditions such as colorectal cancer. Additionally CENPT-related defects have connections to Aurora B kinase a mitotic regulator whose dysfunction associates with cancerous phenotypes.
