RECK also known as Reversion-inducing-cysteine-rich protein with kazal motifs is a 110 kDa membrane-anchored glycoprotein. It plays a role in inhibiting matrix metalloproteinases (MMPs) like MMP-2 and MMP-9 which are involved in extracellular matrix remodeling. RECK is widely expressed in various tissues including the brain heart and muscles. It interacts with other membrane proteins to modulate different cellular functions playing a part in maintaining the structure of tissues.
Biological function summary
RECK supports angiogenesis and tissue integrity by controlling matrix degradation. It is essential for proper vascular development. Although RECK is not part of a bigger complex it exerts its function by partnering with integrins and other membrane components. This interaction helps regulate cell migration and adhesion which are significant for normal development and wound healing processes.
Pathways
RECK engages in the regulation of the extracellular matrix and signaling cascades. It aligns with critical pathways like the Wnt and Rho GTPase pathways. These pathways are important in controlling cellular proliferation and movement. RECK's interaction with proteins like β-catenin and Ras further illustrates its importance in modulating cellular pathways connected to growth and movement.
RECK can affect cancer progression and metastasis. Its downregulation or malfunction is associated with increased tumor invasion and angiogenesis due to uncontrolled MMP activity. RECK's connection with MMP-2 and MMP-9 is significant here as these proteins promote tumor metastasis. Its role is also evident in neurological disorders like cerebral ischemia where defective matrix remodeling can exacerbate tissue damage. Researchers focus on RECK to develop strategies for cancer therapeutics and neuroprotective treatments.
