Follistatin also known as FS or FST is a glycoprotein with an approximate mass of 32 kDa. This protein is part of the follistatin family and plays an important role in various physiological mechanisms. Follistatin synthesizes and secretes in different tissues including the liver gonads and skeletal muscles. It binds to and inhibits members of the transforming growth factor-beta (TGF-β) superfamily particularly activin. This binding prevents activin from interacting with its receptors.
Biological function summary
Follistatin modulates multiple processes such as cell growth differentiation and reproductive physiology. It acts as a binding protein for activin neutralizing its activity. By preventing activin from engaging with its receptor complexes follistatin influences developmental and regenerative processes. Despite not being a part of a transmembrane complex its interaction with activin impacts extracellular interactions.
Pathways
Follistatin plays a significant role in the regulation of the TGF-β signaling pathway and the muscle growth pathway. In the TGF-β pathway it counteracts the signaling of activin which influences cellular homeostasis and inflammatory responses. In the muscle growth pathway follistatin interferes with myostatin another member of the TGF-β family promoting muscle growth. This cross-talk with proteins such as myostatin demonstrates follistatin's influence in muscle regeneration and development.
Follistatin has clear associations with muscle-related and reproductive disorders. It is linked to muscle wasting diseases such as muscular dystrophy due to its inhibitory role on myostatin known to restrain muscle growth. Follistatin abnormalities can also contribute to conditions like polycystic ovary syndrome (PCOS) due to its regulation of reproductive processes. These ties display follistatin's vital interactions with proteins like activin and myostatin highlighting its importance in disease pathways.
