ARHGAP4 also known as Rho GTPase-activating protein 4 is an important regulator in cellular signaling involved in modulating the activity of Rho family GTPases. These proteins play a role in remodelization of the actin cytoskeleton cellular migration and morphogenes. ARHGAP4 has a molecular mass of approximately 113 kDa. It expresses in various tissues with notable expression in the brain spleen and lymphoid organs suggesting its functions in different biological processes and systems.
Biological function summary
ARHGAP4 contributes significantly to the regulation of actin dynamics and cell morphology. This protein is not typically part of a complex but interacts with other proteins to control cell structure and movement. Its activity influences importance in neuronal development and immune response indicating its diverse roles in cellular functions. Moreover it helps in controlling cell motility and adhesion affecting how cells interact with their environments and neighboring cells.
Pathways
ARHGAP4 is intricately involved in the Rho GTPase cycle and the regulation of the actin cytoskeleton pathway. This protein acts as a nexus by inactivating Rho GTPases such as RhoA Rac1 and Cdc42 modulating the signals that lead to cytoskeletal rearrangement. Such interactions find relevance in pathways that orchestrate cellular movement and stabilization significantly impacting pathways that control cell shape and motility.
ARHGAP4 shows associations with neurological disorders and certain cancers. Alterations in its expression or activity are linked to developmental brain abnormalities affecting neuronal migration and connectivity. In cancer its dysregulation may lead to changes in tumor cell motility promoting invasion and metastasis. Proteins such as RhoA and Rac1 which interact with ARHGAP4 are also involved in these diseases highlighting the importance of Rho GTPase signaling pathways in disease pathology.
