ME1 also known as malic enzyme 1 is a cytosolic enzyme that catalyzes the reversible oxidative decarboxylation of malate to pyruvate generating NADPH in the process. This enzyme contributes to cellular lipid and cholesterol biosynthesis. ME1 has an approximate molecular weight of 64 kDa. You can find ME1 expression mainly in tissues with high biosynthetic activities such as liver fat and kidney.
Biological function summary
ME1 plays an important role in regulating the cellular redox state and maintaining metabolic homeostasis. It supplies NADPH which is necessary for fatty acid synthesis and lipid metabolism supporting anabolic processes in cells. ME1 works as a part of a metabolic enzyme complex that coordinates with other enzymes to produce reducing equivalents required for detoxification and biosynthesis. Scientists frequently study its role in lipid synthesis due to its implications in metabolic disorders.
Pathways
ME1 is involved in lipid metabolism and the pentose phosphate pathway where it helps generate reducing power. ME1 interacts with other enzymes like glucose-6-phosphate dehydrogenase which also provides NADPH showing its interconnected role in maintaining cellular redox balance. ME1’s activity supports both pathways by contributing to the production of ribose-5-phosphate and other precursors important for nucleotide biosynthesis and antioxidant defense.
ME1 has links to obesity and cancer where its dysregulation can promote tumorigenesis and metabolic disorders. Studies suggest aberrant ME1 expression may lead to altered lipid biosynthesis contributing to excessive fat accumulation in obesity. ME1 and glutamine synthetase a protein connected in cancer metabolism work together to support cell proliferation through their metabolic pathways highlighting ME1’s potential as a target in metabolic and cancer therapy.
