Product Description
AdvanceBio Sialidase A, 66 kDa. A lower molecular weight version of GK80040 sialidase A for challenging digestions. Enzyme releases α(2-3)-, α(2-6)-, α(2-8), and α(2-9)-linked N-acetylneuraminic acid from oligosaccharides and glycoproteins. Also capable of releasing N-glycolylneuraminic acid. Lower mol wt sialidases may help desialylate glycans if steric hindrance is an issue. If you think a lower mol wt sialidase will help desialylate your glycans, you can also try our 88, 66 and 51 kDa Sialidase A sampler kit (GK80043).

Sialidases are also known as neuraminidases. Specifications: Concentration: ≥5 U/mL Enzyme Source: Recombinant gene from Arthrobacter ureafaciens, expressed in E. coli. Sialidases are also known as neuraminidases. Enzyme Specificity: The enzyme releases a(2,3)-, a(2,6)-, a(2,8)-, and a(2,9)-linked N-acetylneuraminic acid from complex carbohydrates. The initial rate of hydrolysis of a(2,6) linkages is reported to be approximately twice that of a(2,3)-linked sialic acid however, in practice, this kinetic selectivity is of little consequence during extended incubations. Effective digestion of glycolipid substrates is facilitated by addition of a detergent, such as sodium taurodeoxycholate to the incubation. Sialidase A is capable of releasing N-glycolylneuraminic acid (Neu5Gc, NGNA) in addition to N-acetylneuraminic acid (Neu5Ac, NANA) [1], although similarly to other sialidases [2] the activity is lower toward Neu5Gc than Neu5Ac. The enzyme can also be used to remove sialic acid from gangliosides [3], glycosphingolipids (ceramide and oligosaccharide) with sialic acid. May be used for exoglycosidase sequencing with Sialidase S (GK80021), which is specific for a(2,3)-linked sialic acids. Sialidases are also known as neuraminidase. Enzyme Unit Definition: One unit is defined as the amount of enzyme required to catalyze the release of 1 µmole of p-nitrophenol from p-nitrophenyl-α-D-N-acetylneuraminic acid per minute at 37° C, pH 5.5 Molecular Weight: 66 kDa Unit: 1 U Volume: 200 µL pH Optimum: 6 **********The following product description is generated by AI and is for reference only. If you have any questions, please contact customer service.********** AdvanceBio Sialidase A, 66 kDa is a highly purified enzyme designed for efficient and selective removal of terminal sialic acid residues from glycoproteins and other biomolecules. This enzyme cleaves alpha2-3, alpha2-6, and alpha2-8 linked sialic acids, making it ideal for research applications focused on glycosylation analysis, biopharmaceutical development, and quality control of monoclonal antibodies. Suitable for use in deglycosylation workflows, the product contributes to streamlined sample preparation prior to advanced analytical techniques such as HPLC, LC-MS, and capillary electrophoresis. Specifically optimized for compatibility with Agilent's AdvanceBio Glycan Mapping workflows, this sialidase enables accurate profiling and structural characterization of complex glycans, facilitating reliable assessment of critical quality attributes in biologics. Its robust activity and specificity support high-performance sample processing for researchers engaged in detailed glycoprotein analysis. In addition to use with other enzymes in the AdvanceBio glycan enzyme portfolio, this sialidase is recommended for integration with Agilent sample preparation kits, UPLC systems, and LC columns. With rigorous quality assurance testing, it ensures reproducibility and consistency in high-throughput laboratory environments. This product is an essential tool for laboratories seeking to advance glycomics, therapeutic protein characterization, and enhance analytical workflows in biologics research.
