Product Description

AdvanceBio N-Glycanase (PNGase F, EDTA free), ≥2.5 U/mL. A recombinant form of PNGase F (100 mU, EDTA free), releases intact N-glycans by cleaving between the innermost GlcNAc and Asn.  Includes 5x N-glycanase reaction buffer, denaturation solution, detergent solution, 5x N-glycanase Tris reaction buffer (MS applications).

Specifications:
Concentration: 2.5 U/mL
Enzyme Formulation: 20 mM Tris-HCl, 50 mM NaCl, (pH 7.5)
Enzyme Source: Recombinant gene from Elizabethkingia meningoseptica, expressed in E. coli. The source organism was previously known as Chryseobacterium [Flavobacterium] meningosepticum. Enzyme is also known as Endoglycosidase F2, endo-beta-N-acetylglucosaminidase. Enzyme also known as PNGase F, peptide-N-glycosidase F, peptide-N4-(N-acetyl-β-glucosaminyl)asparagine amidase.
Enzyme Specific Activity: ≥10 units/mg
Enzyme Specificity: Cleaves all N-linked complex, hybrid or high mannose oligosaccharides, unless a(1-3) core fucosylated, as in plant glycans. Asparagine must be peptide bonded at both termini. Phosphate, sulfate and sialic acid groups attached to the oligosaccharide do not affect cleavage. Endo F free. Highly concentrated, particularly useful for deglycosylation under native conditions.
Enzyme Unit Definition: One unit is defined as the amount of enzyme required to catalyze the release of N-linked oligosaccharides from 1 µmole of denatured ribonuclease B per minute at 37°C, pH 7.5.
Molecular Weight: ~35,000 Daltons
Unit: 100 mU
Volume: 40 µL
pH Optimum: 8.6
pH Range: 7.5-9.5

AdvanceBio N-Glycanase is a highly purified recombinant PNGase F enzyme formulated without EDTA, delivering an activity of at least 2.5 units per milliliter. This enzyme efficiently cleaves N-linked oligosaccharides from glycoproteins, making it an essential tool for streamlined glycan profiling, characterization, and quality control in biopharmaceutical development and research.

Optimized for analytical workflows, this reagent enables rapid and complete deglycosylation of both native and denatured glycoproteins under mild conditions, helping preserve sensitive protein structures. It is particularly effective in preparing samples for downstream analysis by techniques such as HILIC-UPLC, LC-MS, and CE, supporting both qualitative and quantitative glycan analysis.

AdvanceBio N-Glycanase is fully compatible with Agilent’s AdvanceBio Glycan Mapping solutions, including sample preparation kits, glycan labeling reagents, and HPLC columns, providing a seamless integration into established glycomics protocols. It is suited for a broad range of research applications, from monoclonal antibody characterization to glycoprotein biomarker studies and biosimilar development. Free of EDTA, this formulation minimizes potential interference with downstream mass spectrometry and enzymatic workflows, supporting the high sensitivity and accuracy required for advanced glycan analysis. This product is ideal for researchers seeking reliable, high-efficiency glycan release for robust analytical results.
