Product Description

AdvanceBio N-Glycanase-ultra (PNGase F, EDTA free), ≥10 U/mL. A recombinant form of PNGase F, releases intact N-glycans by cleaving between the innermost GlcNAc and Asn. The same formulation provided in GlykoPrep, ≥ 10 U/mL and EDTA-free. Supplied as 5 x 400 mU vials of GKE-5020B. Includes 5x N-glycanase reaction buffer, denaturation solution, detergent solution.

Specifications:
Concentration: 10 U/mL
Enzyme Applications: To obtain efficient deglycosylation of glycoprotein substrates under non-denaturing conditions, it is necessary to use a higher starting concentration of enzyme. N-Glycanase PLUS and ULTRA (EDTA-Free) are supplied at = 10 U/ml, and are recommended for all applications requiring deglycosylation of glycoproteins in the absence of denaturants. The high activity also allows smaller reaction volumes and shorter reaction times to be explored.
Enzyme Formulation: 20 mM Tris-HCl, 50 mM NaCl (pH 7.5, EDTA-free formulation).
Enzyme Source: Recombinant gene from Elizabethkingia meningoseptica, expressed in E. coli. The source organism was previously known as Chryseobacterium [Flavobacterium] meningosepticum. Enzyme is also known as Endoglycosidase F2, endo-beta-N-acetylglucosaminidase. Enzyme also known as PNGase F, peptide-N-glycosidase F, peptide-N4-(N-acetyl-β-glucosaminyl)asparagine amidase.
Enzyme Specific Activity: ≥10 units/mg
Enzyme Specificity: Cleaves all N-linked complex, hybrid or high mannose oligosaccharides, unless a(1-3) core fucosylated, as in plant glycans. Asparagine must be peptide bonded at both termini. Phosphate, sulfate and sialic, acid groups attached to the oligosaccharide do not affect cleavage. Endo F free. Highly concentrated, particularly useful for deglycosylation under native conditions.
Enzyme Unit Definition: One unit is defined as the amount of enzyme required to catalyze the release of N-linked oligosaccharides from 1 µmole of denatured ribonuclease B per minute at 37°C, pH 7.5.
Molecular Weight: ~35,000 Daltons
Unit: 2000 mU
Volume: 400 µL
pH Optimum: 8.6
pH Range: 7.5-9.5

AdvanceBio N-Glycanase-ultra is a highly purified recombinant Peptide-N-Glycosidase F designed for efficient and reliable deglycosylation of N-linked glycans from glycoproteins. Free from EDTA, this enzyme preparation is supplied at a concentration of at least 10 units per milliliter, ensuring optimal activity for sensitive downstream analyses. Its robust formulation allows precise and rapid release of intact N-glycans under gentle conditions, making it ideal for sample preparation in biopharmaceutical characterization, quality control, and glycosylation profiling.

The product is compatible with a range of Agilent analytical platforms including LC, LC-MS, and capillary electrophoresis, ensuring seamless integration into established glycan analysis workflows. It is particularly well suited for use with AdvanceBio Glycan Mapping columns and relevant sample preparation kits, supporting high-resolution analysis and reproducible results.

AdvanceBio N-Glycanase-ultra is recommended for researchers focused on monoclonal antibody development, biosimilar studies, vaccine research, or glycoprotein therapeutics, where precise N-glycan release is critical. The absence of EDTA reduces the risk of metal cation interference, accommodating applications with metal-dependent enzymes or sensitive detection systems. This reliable reagent streamlines glycan sample processing, delivering consistent performance for demanding research and analytical needs.
