The Siglec-H is a member of CD33-related Siglec family which mostly expressed in the innate immune system. Siglec-H is reported to be specifically expressed on plasmacytoid dendritic cells. Siglec-H depends on adaptor protein DAP 12 (DNAX-activating protein of 12 kD) for surface expression and signal transduction. Cross-linking with this antibody can selectively inhibit interferon-alpha production by IPCs following TLR9 ligation.

Application: FC - Quality tested
Recommended Usage: Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25 µg per 106 cells in 100 µl volume. Distribution: Siglec-H is expressed exclusively on natural interferon-producing cells (IPCs), also called plasmacytoid dendritic cells in spleen, blood and lymph nodes. The IPCs are characterized by a complex phenotype (CD11c+, Ly-6C+, B220+, CD11b-, CD8+/-).
Function: Siglec-H associates with adaptor protein DAP12 for cell signaling, which reduces type 1 IFN response of IPC to CpG.
