Cell-Vive™ GMP CD3/CD28 Human T Cell Activation Beads allow for convenient in vitro or ex vivo activation of human T cells without the need for antigen-presenting cells. Human T cells are stimulated with anti-CD3 and anti-CD28 antibodies coated on superparamagnetic microbeads. Simply add the beads directly to isolated T cells or to PBMCs at a 1:1 bead-to-cell ratio for robust T cell activation. Excess antibody-coated beads can be removed from the cell suspension by the use of a magnetic separator (some residual beads may be retained in the cell suspension). Some of the downstream applications include functional assays, gene expression analysis, and phenotypic characterization. Recommended Usage: GMP CD3/CD28 Human T Cell activation beads should be plated with cells at a 1:1 bead-to-cell ratio.
Application Notes: This product is designed for the in vitro or ex vivo expansion of human T cells. Cell-Vive™ GMP CD3/CD28 Human T Cell Activation Beads are a ready to use solution. There is no need to dilute the product prior to use in cell culture. Suggested culture protocol: Prepare T Cell Complete Media (TCCM) using sterile, filtered IMDM with 5% final concentration of serum and 200 IU/mL of human IL-2. Prepare cells at a concentration of 1 x 106 per mL of TCCM. Wash 25 µL of Cell-Vive™ GMP CD3/CD28 Human T Cell Activation Beads to a 5 mL polypropylene tube with 1 mL TCCM. If using a 1:1 bead-to-cell ratio, wash 25 µL of beads per sample. Adjust the volume of beads depending on final bead-to-cell ratio. For isolated T cells, we recommend using a lower bead-to-cell ratio. 1:3 or 1:4 is recommended. Apply tube containing beads to a magnet (MojoSort magnet, cat#: 480019) for 2 minutes. Decant supernatant and remove tube from magnet. Resuspend beads in 1 mL of cell suspension and plate in a 12 well cell culture plate. Allow cells to expand at 37°C and 5% CO2. At Day 4 and Day 7, count cells and resuspend at 0.5 x 106 cells per mL. Harvest cells at Day 10 or Day 11 for further analysis. This product was optimized on isolated CD3+ T cells and PBMCs. Alternate sources of T cells may require protocol optimization.
