Biolegend, 423001, Stop Solution for TMB Substrate, 60ml

Biolegend’s Stop Solution for TMB Substrate is specifically formulated to terminate the HRP/TMB reaction and stabilizing the color development for ELISA applications. TMB is a chromogenic substrate used in ELISA. TMB can act as a proton donor for the reduction of hydrogen peroxide to water by peroxidase enzymes such as HRP. The resulting diimine causes the solution to take on a blue color, and this color change can be read at 650 nm. The reaction can be stopped by addition of stop reagent, and the color turns from blue to yellow with 2-3 fold increased absorbance values at 450 nm.

Preparation: This solution is ready to use and requires no dilution.

Application Notes: Each lot of Stop Solution for TMB (3,3'5,5'-tetramethylbenzidine) Substrate has been tested by ELISA. The TMB stop solution will stop TMB and HRP (horseradish peroxidase) reaction by adding the same volume as the TMB substrate added to each well. Addition of stop solution changes the color from blue to yellow. Read absorbance at 450 nm within 60 minutes. If the reader can read at 570 nm, the absorbance at 570 nm can be subtracted from the absorbance at 450 nm.
