The Swift-Click™ Green EdU Kit allows the rapid fluorescent detection of incorporated nucleoside analog in proliferating cells for cell cycle analysis. The staining procedure consists of labeling cells through incubation with nucleoside analog 5-ethynyl-2´-deoxyuridine (EdU), followed by cell fixation and permeabilization using our True-Nuclear™ Transcription Factor Buffer Set, and fluorescent EdU labeling. Fluorescent EdU labeling is catalyzed by Click chemistry, a Nobel Prize-recognized chemical reaction that rapidly links fluorophore-azide molecules to the alkyne group present in EdU in the presence of Cu 2+ ions. This method bypasses radioactivity use, as well as long and harsh permeabilization and chromatin denaturation steps, allowing for a faster, simpler-to-follow procedure for cell cycle analysis.
