Neurogranin (NRGN) is the main post-synaptic protein kinase substrate that binds to calmodulin (CaM) in the absence of calcium. NRGN’s size (78 amino acids, 7.6 kD) allows it to pass through a damaged blood–brain barrier, making it a biomarker in chronic neurodegenerative diseases and potentially for acute brain injury. Measurement of serum NRGN levels could support the early diagnosis of central nervous infections. Neurogranin in CSF has been implicated as a possible marker for synaptic degeneration in Alzheimer’s disease. NRGN function has been implicated in learning and memory. It is suggested that the NRGN is a direct target for thyroid hormone in the human brain and the control of expression of this gene could underlay many of the consequences of hypothyroidism on mental states during development, as well as in adult subjects.
