Angiopoietin-1 encodes a 498 amino acid polypeptide with a molecular weight of 57 kDa. There are now four identified Angiopoietins: Angiopoietin-1, Angiopoietin-2, Angiotpoietin-3, and Angiopoietin-4. Angiopoietin-1 is a largely endothelial-specific growth factor/ligand/agonist that acts through the vascular tyrosine kinase receptor Tie-2. Angiopoietin-1 activates Tie-2 to initiate Angiopoietin/Tie-2 pathway. Signaling by Angiopoietin-1 promotes vascular endothelial cell survival and the sprouting and reorganizing blood vessels, as well as inhibiting activation of the vascular endothelial barrier to reduce leakage and leucocyte migration into tissues. Structurally, Angiopoietin-1 has an N-terminal super clustering domain, a central coiled domain, a linker region, and a C-terminal fibrinogen-related domain responsible for the binding between the ligand and receptor (Tie-2). Angiopoietin-1 can form dimers, trimers, and tetramers. Angiopoietin-1 can form higher order multimers through its super clustering domain. However, not all the structures can interact with the tyrosine kinase receptor. The receptor can only be activated at the tetramer level.
