CD28H, initially called TMIGD2 (transmembrane and immunoglobulin domain containing 2), is a single transmembrane protein with one immunoglobulin variable-like (IgV) domain, and a long intracellular domain. CD28H shares approximately 10% homology with CD28, CTLA, ICOS and PD-1. CD28H is conserved in zebra fish, cow, guinea pig, and chimpanzee but the coding gene is not present in mouse and rat. The ligands for CD28H are B7-H7 and CD28H. B7-H7 orthologues do not exist in mice and rats. B7-H7 is expressed in multiple human cancers including osteosarcomas, oral, bladder, intrahepatic, and colorectal cancers. Interaction of APC or B7-H7 positive tumor cells with primary NK and naïve T cells expressing CD28H induces activation of naïve NK and T cells. CD28H/B7- H7 interaction promotes T cell proliferation and cytokine secretion including IFN-γ, IL-5, IL-10, and IL-17. The interaction of B7-H7 and CD28H also promotes NK cell degranulation. After activation of T and NK cell, expression of CD28H progressively declines. Interaction between B7-H7 and KIR3DL3 may produce antagonistic effects compared to effects elicited from CD28H/B7-H7 interaction. When activated KIR3DL3 positive T and NK cells interact with APC or B7-H7 positive cancer cells, T cell proliferation, cytokine production, and NK cell activation are inhibited.

Source: Recombinant human TMIGD2 (CD28H)-Fc Chimera, amino acid sequence containing Leu23-Gly150 (Accession No. Q96BF3) was expressed in 293E cells. The carboxy-terminus contains SR-IEGRMD-hIgG1 (Pro100-Lys330)-Fc tag.
Molecular Mass: The 367 amino acid recombinant protein has a predicted molecular mass of approximately 40.8 kD. The DTT-reduced and non-reduced proteins migrate at approximately 60 -70 kD and 100 -110 kD by SDS-PAGE, respectively. The predicted N-terminal amino acid is Leu.
Purity: > 95%, as determined by Coomassie stained SDS-PAGE.
Formulation: 0.22 µm filtered protein solution is in PBS, pH 7.2
Endotoxin Level: Less than 0.1 EU per µg cytokine as determined by the LAL method.
Concentration: 25 µg size is bottled at 200 µg/mL. 100 µg size and larger sizes are lot-specific and bottled at the concentration indicated on the vial. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
Storage & Handling: Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.
Activity: When recombinant human TIMIGD2 (CD28H)-Fc is immobilized at 2 µg/mL, recombinant human B7-H7 binds in a dose-dependent manner. HRP anti-His tag (Cat. No. 652503) was used to detect the binding. The ED50 for this effect is 1.5 – 9 µg/mL.

A: The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.
Q: Have your recombinants been tested for stability?
A: Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.
