Product Description

Signal Regulatory Proteins (SIRPs) are a family of transmembrane glycoproteins with extracellular IgG domains. The extracellular regions of the SIRPs are comprised of three Ig-like loops; the proximal loop containing two IgC domains while the distal membrane loop contains one IgV domain. While these extracellular domains are fairly similar among SIRPs, their transmembrane and cytoplasmic domains differentiate them from one another. Signal regulatory protein α (SIRPα) is highly expressed on macrophages and dendritic cells. Interaction of the extracellular region of SIRPα with its ligand CD47 elicits signaling and SIRPα phosphorylation. CD47-SIRPα signaling results in the recruitment and activation of the tyrosine phosphatases SHP-1 and SHP-2, which regulate cellular functions in a negative fashion, including the regulation of leukocyte adhesion and transmigration, T cell activation, macrophage fusion and phagocytosis. In addition to its role in "self-signaling", SIRPα also functions as a scaffold for membrane-bound signaling molecules. Interaction of CD47 on tumor cells with SIRPα on tumor-associated macrophages (TAMs) constitutes an innate immune checkpoint that interferes with the detection and phagocytosis. SIRPα V1, SIRPα V2, and SIRPα V8 are the main SIRPα variants in human. hSIRPα V1 is dominant among Europeans, Africans, and South Asians while hSIRPα V2 dominates east Asians. Sequence alignment of hSIRPα V1, hSIRPα V2, and hSIRPα V8 proteins demonstrated the differences within the CD47-binding extracellular Ig-like V-type (IgV) domain.

Source: Biotinylated human SIRPα V2 amino acids Glu31-Arg369 (Accession # P78324) with a C-terminal His and Avi tag was expressed in CHO cells. Human SIRPα V2-Avi tag was site-specifically biotinylated by enzyme BirA.
Molecular Mass: The 360 amino acid recombinant protein has a predicted molecular mass of approximately 39.7 kD. The DTT-reduced and non-reduced protein migrate at approximately 50-60 kD in SDS-PAGE.

Purity: > 95%, as determined by Coomassie stained SDS-PAGE.

Concentration: 25 µg size is bottled at 200 µg/mL. 100 µg size and larger sizes are lot-specific and bottled at the concentration indicated on the vial. To obtain lot-specific concentration and expiration, please enter the lot number in our Certificate of Analysis online tool.
Storage & Handling: Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at - 20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 μg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.
Activity: Immobilized biotinylated recombinant human SIRPα V2 supports adhesion of human red blood cells in dose-dependent manner. ED50 of this effect is 1.2 – 9.6 µg/mL.
