Gp130 (glycoprotein 130), also known as CD130, IL6ST (IL-6-receptor-associated signal transducer), or common β chain, is a type I transmembrane glycoprotein consisting of 597 amino acid (aa) extracellular domain, one Ig-like domain, five fibronectin type-III repeats, a short transmembrane domain, and 277 amino acids intracellular domain. CD130 is expressed on T cells, activated B cells, monocytes, plasma cells, endothelial, and epithelial cells. Gp130 is a common subunit shared by a variety of functional cytokine receptors within the IL-6 and IL-12 family, including IL-6, IL-11, leukemia inhibitory factor (LIF), Oncostatin M (OSM), ciliary neurotrophic factor (CNTF), cardiotrophin-1 (CT-1), novel neutrophin-1/B cell stimulating factor-3 or cardiotrophin-like cytokine (CLC), Neuropoietin (NP), IL-27, IL-35, and IL-39. Gp130 is utilized as the signal-transducing molecule by these receptor complexes for initiating signal transmission. Binding of IL-6 to IL-6 R (IL-11 R) induces gp130 homodimerization to form the high-binding monomeric four-helix bundle complex, which activates the intracellular Janus Kinase/Signal Transducer and Activator of Transcription (JAK/STAT) signaling pathways. Gp130 promotes mainly pro-inflammatory immune responses, prevents apoptosis and inhibits of regulatory T cell (Treg) differentiation, regulate hematopoiesis, and bone metabolism. It also has been shown to have a role in embryonic development and is essential for the survival of motor and sensory neurons and for the differentiation of astrocytes.

Source: Mouse gp130, amino acid (Gln23-Glu617) (Accession: # Q00560.2), with a linker (AAANSSLGS) and a C-terminal human IgG1 (Pro100-Lys330), followed by a 6His tag, was expressed in CHO cells.
Molecular Mass: The 841 amino acid recombinant protein has a predicted molecular mass of approximately 94 kD. The DTT-reduced and non-reduced proteins migrate at approximately 115 - 130 kD and 240 kD by SDS-PAGE. The predicted N-terminal amino acid is Gln.
Purity: > 95%, as determined by Coomassie stained SDS-PAGE.

Endotoxin Level: Less than 0.1 EU per µg cytokine as determined by the LAL method.

Storage & Handling: Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin the vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.
Activity: Recombinant mouse gp130 inhibits the proliferation of mouse 7TD1 cells induced by recombinant human IL-6 in the presence of Recombinant Human IL-6Rα (Cat. No. 751502). The ED50 for this effect is 0.4 – 4 ng/mL.

A: The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.
Q: Have your recombinants been tested for stability?
