SMAD2 is a potent regulator of different cellular processes such as apoptosis, differentiation, and cell proliferation. SMAD2 is associated with SARA protein and the TGF-B receptor. SMAD2 gets phosphorylated by the TGF-B receptor in response to TGF-B signal, which induces its dissociation from SARA and association with SMAD4 followed by translocation into the nucleus. In the nucleus, the SMAD2/SMAD4 protein participates in gene regulation by forming a transcription repressor complex in association with other cofactors.

Recommended Usage: Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.5 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.
Application Notes: Since SMAD2 shares a high percentage of identity with SMAD3, this clone recognizes both SMAD2 and SMAD3, which can be resolved by Western Blot.

Structure: SMAD2 is a 467 amino acid protein with a predicted molecular weight of 52 kD. SMAD2 is very similar in sequence and structure to SMAD3.
Distribution: Non-phosphorylated SMAD2 is present in the cellular cytoplasm close to the TGF-B receptor. Phosphorylated SMAD2 is present in the cellular nucleus bind to target promotor sequences.
Function: The function of SMAD2 is mainly as a gene expression regulator. SMAD2 acts as a sequence specific transcriptional repressor.
Interaction: Non-phosphorylated SMAD2 interacts with SARA (SMAD anchor for receptor activation) protein. Phosphorylated SMAD2 interacts with SMAD4 and other members of the transcription repressor complex.
