MALT1 is a ubiquitously expressed protease that is localized in the cytoplasm as a catalytically inactive BCL10-bound proenzyme. MALT1 acts downstream of oligomeric CARMA/CARD-BCL10-MALT1 (CBM) complexes to regulate various functions in which MALT1 serves a dual purpose. Firstly, MALT1 is a scaffolding protein that plays an integral role in the regulation of the NF-κB and MAP kinase p38 pathways. In addition to serving as a scaffolding protein, MALT1 also possesses protease activity which promotes lymphocyte activation by cleavage of signaling proteins. Furthermore, MALT1 possesses a caspase-like domain with two conserved residues, cysteine 464 and histidine 415, which act as catalytic mechanisms in caspases and cysteine proteases ultimately contributing to inflammation, cell death, and cell proliferation.

Recommended Usage: Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 5.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.25 - 5.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
Application Notes: For ICC, we recommend fixation with 4%PFA, followed by permeabilization with 0.5% Triton-X or 100% ice-cold methanol. We do not recommend methanol fixation. This antibody is not suitable for WB due to poor target selectivity.
